Tutorial - Manual Serial Dilution
Introduction¶
In this tutorial, you will perform a dilution by row from a 96-well plate with a single column of filled wells (source concentration = 10 mM, Dilution Factor = 2) manually.

These should exist by default in the Sample application.
Items¶
Containers and locations created from Discngine Sample’s automation will be used:
- A plate with a single column of filled wells (easily created with the workflow
Plate creation from vials) corresponding to column 1. Automation is running during installation if “example_dataset” is set to true in the configuration file.
Workflow¶
Job Initialization¶
Create a new job and select the Plate Serial Dilution workflow.
Requesting¶
- Select Greiner PP96V as the Target Plate Type
- Specify Request from Text
- Enter the name of your plate (Prerequisite: a plate with empty wells except in its first column). In this example:
P000000001. - Click
Add IDsto confirm. The vials are listed in the added requests. - Click
Next
Task Requesting¶
- Dilution Template = Manual Layout
- Dilution by = Row
- Initial Column Index = 1
- Target Colum Index = 2
- Dilution factor = 2
- Solvent = DMSO
- Click
Add Task. The Source Column Index and the Target Column Index are automatically updated. ClickAdd Taskuntil Target Column Index = 11. - Click

Sourcing¶
The Sourcing page is used to check that the requested operations are compatible with the source plate (information in the Sourced column) and available.

Processing¶
For processing, requested tasks are translated into a worklist that you can feed to your robotic equipment.
- Select the Standard (Well) (;) equipment and set File Order to By Request
- Click
Generate worklist
A worklist file appears in the Worklist Generator area. It contains the requested tasks to execute. The Worklist Parser area expects a file containing the information about the tasks that were actually completed. By default, the generated file is set in the Worklist Parser. For this tutorial, we assume that every task was completed successfully, so we’ll keep this default file.

Nextto propagate those changes in the system. The tasks are processed in the Sample application.
Storing¶
The Storing step allows you to assign a location to the items involved in the job. In this workflow, as the serial dilution is done within the plate, the input plate is the output plate.
Click on the barcode to see the plate content. Samples are filled from column 1 to column 11.

Nextto continue. Here, we won’t change the input plate's location. You can increment the Thawing Cycle count of the input plates if necessary.
Pipe & Close¶
The Pipe & Close step provides a summary page where you can review the job and optionally pipe it to another workflow.
You can also pipe this job to another one, allowing you to immediately start a new job using the output or the input plates from this one.
For this tutorial, click Close & Quit to finish the job.
